Evaluación in vitro de la actividad inhibidora de fracciones de Passiflora edulis Sims. (Passifloracea) sobre la enzima convertidora de angiotensina

Currently, the prevalence of hypertensive patients is increasing due to psychosocial, behavioral, inherited factors, among others. The use of drugs could lead to the appearance of undesirable effects during the treatment of the disease. Traditional medicine with plants provides alternatives to show potential therapeutic candidates. There is evidence of the effect of Passiflora edulis Sims juice. on the angiotensin-converting enzyme (ACE). In this work, 4 large fractions were obtained by column chromatography using DIAION HP-20 from the juice
of Passiflora edulis Sims., The separation was monitored by U.V. vis analysis and CCD chromatography; the activity of the RCT in blood serum was evaluated, using the Ronca- Testoni method (74), modified by the Biochemistry and Genetics laboratory of the University
of Quindio. We included 5 healthy volunteers without diagnosis of hypertension and with informed consent. Fractions A and B were evaluated at 4 concentrations (10 μg / mL, 20 μg /mL, 50 μg / mL, 100 μg / mL). Both fractions showed inhibitory activity in vitro on RCTs similar
to the drug Captopril (ActA = 40.60 + 22.07, ActB = 44.74 + 24.92, Capt = 41.79 + 32.03, White = 66.33 +23.6 μg / mL). The percentage of ACE inhibition activity for the concentrations was approximately 38.79%. The values of the ACE inhibitory activity due to the effect of the
fraction at different concentrations were not statistically significant. It was evidenced by FTIR spectroscopy the existence of metabolites with characteristics of aromatic carboxylic compounds of unsaturated and aliphatic chains, possibly phenolic esters, as major compounds in both fractions. It is proposed that the inhibition of the enzyme may be
mediated by the interactions of the functional groups with the zinc atom of the metalloproteinase and the formation of hydrogen bonds between the phenolic metabolites and the amino acids of the catalytic site. The metabolites of these fractions could act as adjuvants by having an enzymatic activity close to that of the control medication. Actualmente, la prevalencia de hipertensos se mantiene en aumento por factores psicosociales, conductuales, heredados, entre otros. El uso de fármacos podría conllevar a la aparición de efectos indeseables durante el tratamiento de la enfermedad. La medicina tradicional con plantas provee de alternativas para evidenciar posibles candidatos terapéuticos. Se tiene evidencia del efecto del jugo de Passiflora edulis Sims sobre la enzima convertidora de angiotensina (ECA). En este trabajo se obtuvieron 4 grandes fracciones por cromatografía en columna usando DIAION HP-20 a partir del jugo de Passiflora edulis Sims., la separación fue monitoreada por análisis U.V vis y cromatografía CCD; se evaluó la actividad de la ECA en suero sanguíneo, usando el método de Ronca–Testoni (74), modificado por el laboratorio de Bioquímica y Genética de la Universidad del Quindio. Se incluyeron 5 voluntarios sanos sin diagnóstico de hipertensión y con consentimiento informado. Se evaluaron las fracciones A y B, a 4 concentraciones (10 mg/mL, 20 mg/mL, 50 mg/mL, 100 mg/mL). Ambas fracciones presentaron actividad inhibitoria in vitro sobre ECA similar al medicamento Captopril (ActA=40,60+22,07; ActB=44,74+24,92; Capt=41,79+32,03; Blanco=66,33+23,6 mg/mL). El porcentaje de actividad de inhibición de ECA para las concentraciones fue aproximadamente del 38,79%. Los valores de la actividad inhibidora de ECA por efecto de la fracción a diferentes concentraciones no fueron estadísticamente significativos. Se evidencio por espectroscopia FTIR la existencia de metabolitos con características de compuestos carboxilicos aromáticos de cadenas insaturadas y alifáticas, posiblemente esteres fenólicos, como compuestos mayoritarios en ambas fracciones. Se propone que la inhibición de la enzima puede estar mediada por las interacciones de los grupos funcionales con el átomo de zinc de la metaloproteinasa y la formación de puentes de hidrogeno entre los metabolitos fenólicos y los aminoácidos del sitio catalítico. Los metabolitos de estas fracciones podrían actuar como coadyuvantes al tener una actividad enzimática cercana a la del medicamento control.
